Comision de seminarios Resumen: Trisomy 21 is consistently associated with neurodevelopmental alterations in individuals with Down syndrome (DS) and confers a very high risk of early onset Alzheimer disease (AD). Also, it is accepted that trisomy for the APP gene on chromosome 21 is a major factor underlying AD in DS. However, the physiological role of APP remains controversial, and it is unclear the impact of increase dosage of APP during neurodevelopment in DS. To unravel the complex biology of APP and its potential role in DS neurodevelopment, we engineered trisomy 21 iPS cell lines using CRISPR/Cas9 and produced isogenic lines that vary in the gene dosage of APP. Our results indicate a reduction in the capability of the differentiation of human neuroprogenitor cell (NPC) to neurons and neurite outgrowth in the cells with dosage reduction of APP compared with the trisomy’s isogenic one. Moreover, Compound E an inhibitor of secretase which is involve in the processing of APP and Notch receptor, restores the capability of APP depleted DS-iPSC to differentiate to neuron. This work reveals the importance of APP as a modulator of differentiation and maturation of neurons in DS and the participation/implication of secretase in the signaling of APP and Notch receptor. -- *Dra.
